Plainly and simply, no. You would have to cut a second hole on the button side of the cuff and still it would not fit right due to the button disturbing the cuff-link.

Also if the cuff has a round or mitred profile it would look off with cuff-links.

You could, if the tailer knows how (it's a kind of specific alteration). If you have a local tailor who makes shirts, they can handle it.

Barrel cuffs can be converted to single French cuffs or convertible cuffs, or the cuffs can just be replaced (usually that involves turning the shirt into a contrast cuff & collar shirt, since an exact color match is impossible, so the tailor just installs white cuffs and collar... depends on the color of your dress shirt).

Depending on circumstances, it's probably a $20-30 job? Worth considering just buying another shirt, one with native French cuffs?

When the buttonhole is cut on the other side, the button is removed so it doesn't get in the way.

A rounded cuff is fine with a cufflink. It's a traditional look for a single link cuff. The only problem is that the cuff will likely be too soft for cufflinks. Single link cuffs are generally stiff. Double cuffs are soft, but the double layer helps support the cufflinks.

For Christmas, I got a dress shirt and some cufflinks. The shirt is just a standard barrel cuff shirt. Is there a way I can have an alterations tailor make the cuff into a hybrid cuff so I can wear it with cufflinks?
The best, and easiest, thing for you to do is to buy a shirt with French cuffs. Messing with the cuffs on this one may result in ruining the shirt and you losing money. Just enjoy this shirt with its barrel cuffs and get another shirt for the cufflinks.
